大五月花號是一款進行四輪的遊戲，適合2-6名玩家。
每輪遊戲代表一季，依序為春、夏、秋、冬。

每名玩家擁有一張「家園板」並帶領一支由藍、紅及黃三色隨機組成的八名工人團隊。
派遣顏色相符的工人競標圖板，以擴大自己的村莊，亦可派遣到自己的村莊、其他玩家的村莊或競標中的圖板上獲得技能、其他工人、分數或生產資源。
不只可以使用自己的圖板，也可以使用其他玩家村莊中的圖板，甚至是正在進行競標的新圖板。
在春、夏及秋季，會有更多的工人登上大五月花號及她的姐妹艦，這些工人擁有生產鐵、石材及木材等重要資源的技能。
在冬季，玩家選擇村莊板進行競標，每張圖板會依資源、技能及工人而提供不同的分數。
大五月花號將帶給玩家許多不同的挑戰，且每次遊戲都不盡相同，因為村莊板隨機出現為遊戲帶來千變萬化的風貌。
遊戲中，玩家要隨時保持警覺，當機會來臨時，善用自己擁有的資源及利用運輸，以提升能力、獲得技能及增加工人。
遊戲結束時，得分最高的玩家獲勝。
